Study on the effect and mechanism of quercetin on type Ⅲ chronic prostatitis/pelvic pain syndrome rats

摘要: 目的:探究槲皮素对Ⅲ型慢性前列腺炎/骨盆疼痛综合征模型大鼠作用及机制。方法:将大鼠随机分为对照组、模型组、槲皮素低剂量组(100mg/kg)和槲皮素高剂量组(200mg/kg),复制Ⅲ型慢性前列腺炎/骨盆疼痛综合征模型。病理检查前列腺,测定机械性痛阈,Western blotting检测前列腺中p38和环氧合酶-2(COX-2)水平,ELISA检测血清中核转录因子-κB (NF-κB)、肿瘤坏死因子-α(TNF-α)水平。结果:槲皮素低剂量组较模型组前列腺组织病理有所改善但不显著,槲皮素高剂量组较模型组前列腺组织病理有显著改善。槲皮素低剂量组的机械痛阈值高于模型组,但两组比较差异无统计学意义(P>0.05),槲皮素高剂量组的机械痛阈值显著性高于模型组(P<0.05);槲皮素低剂量组血清中NF-κB和TNF-α水平、前列腺组织中p38和COX-2水平低于模型组,但两组比较差异无统计学意义(P>0.05),槲皮素高剂量组的血清中NF-κB和TNF-α、前列腺组织中p38和COX-2水平显著低于模型组(P<0.05)。结论:槲皮素对Ⅲ型慢性前列腺炎/骨盆疼痛综合征模型大鼠的前列腺病理和痛觉敏感性均有改善作用,其作用机制可能为降低多种炎性因子的表达,抑制炎症反应和痛觉敏感性。Abstract: Objective: To explore the effect and mechanism of quercetin on typeⅢ chronic prostatitis/pelvic pain syndrome rats.Method: The rats were randomly divided into the control group, the model group, the quercetin low dose group (100 mg/kg) and the quercetin high dose group (200 mg/kg).TypeⅢchronic prostatitis/pelvic pain syndrome model was made.Pathological examination of prostate was done, mechanical pain threshold was examined, western blotting was used to detect the levels of p38 and COX-2 in the prostate, and ELISA was used to detect the levels of NF-κB and TNF-αin serum.Result: Compared with the model group, the pathological changes of prostate tissue in the quercetin low dose group were a little improved, but the pathological changes of prostate tissue in the quercetin high dose group were significantly improved compared with the model group.The mechanical pain threshold of the quercetin low dose group was higher than that of the model group, but there was no significant difference between the two groups (P>0.05).The mechanical pain threshold of the quercetin high dose group was significantly higher than that of the model group (P<0.05).The serum levels of NF-κB and TNF-α, p38 and COX-2 in prostate tissue were lower in the quercetin low dose group than those in the model group, but there was no significant difference between the two groups (P>0.05), The serum levels of NF-κB and TNF-α, p38 and COX-2 in prostate tissue were significantly lower in the quercetin high dose group than those in the model group (P<0.05).Conclusion: Quercetin can improve typeⅢ chronic prostatitis/pelvic pain and pain sensitivity in rat models, and reducing expression of various inflammatory cytokines and inhibiting inflammation and pain sensitivity may be the mechanisms.
