Expression of PTGDS in prostate cancer and its correlation with AR

  • 目的:挖掘前列腺癌进展相关基因并探讨其在前列腺癌中的表达以及与临床参数之间的关系,预测其对前列腺癌进展和预后的影响。方法:通过Oncomine和GEO数据库筛选前列腺癌组织和癌旁组织的测序数据;运用R语言分析数据得到差异表达基因,取交集后筛选出前列腺素D2合成酶(PTGDS);分析其与前列腺癌临床参数以及与雄激素受体(AR)的相关性,通过基因富集分析(GSEA)研究PTGDS在肿瘤中可能的作用和功能。结果:PTGDS在前列腺癌组织中较正常组织呈现相对低表达;免疫组化进一步证实这种表达模式;PTGDS与多种临床参数相关,PTGDS低表达预示更差的预后;GSEA结果提示PTGDS与前列腺癌和AR密切相关,PTGDS表达水平与AR呈明显负相关。结论:PTGDS在前列腺癌中低表达,与前列腺癌的发生发展及多个临床参数相关,且与AR呈负相关,有潜力成为前列腺癌诊断和预后的新的生物标记物。
