Expression and significance of Notch1、Notch2 and c-Myc in clear cell renal cell carcinoma

摘要: 目的:探讨Notch信号受体Notch1、Notch2及c-Myc在肾透明细胞癌中的表达与意义。方法:通过实时定量PCR及western-blot方法检测27例肾透明细胞癌及瘤旁组织中Notch1、Notch2及c-Myc的表达水平,并采用相关性分析检验Notch1、Notch2的表达与c-Myc的表达相关性。结果:Notch1、Notch2及c-Myc在肾癌组织中表达水平明显高于相应的癌旁组织(P<0.001),相关性分析表明在肿瘤组织中c-Myc与Notch2的mRNA水平呈正相关(r=0.629,P<0.001),而与Notch1的mRNA水平无相关(P=0.389)。结论:Notch信号通路及c-Myc的异常高表达可能与肾透明细胞癌的发生发展有关,而且c-Myc可能作为Notch信号通路的下游靶基因,发挥促癌的作用。Abstract: Objective:To investigate the expression and significance of Notch1, Notch2 and c-Myc in clear cell renal cell carcinoma (ccRCC). Method:Message RNA and protein levels of Notch1, Notch2 and c-Myc were detected in 27 ccRCC tissue samples and their corresponding adjacent normal tissue samples by real-time PCR and western-blot, respectively. Correlation analyses were utilized between mRNA expressions of Notch1, Notch2 and c-Myc. Result:Notch1, Notch2 and c-Myc were all upregulated in ccRCC samples relative to adjacent normal tissue samples (P<0.001). Correlation analyses showed that c-Myc mRNA expression was closes correlated with Notch2 mRNA expression in ccRCC (r=0.629, P<0.001), but not with Notch1 mRNA expression (P=0.389).Conclusion:Up-regulated Notch1, Notch2 and c-Myc may associate with ccRCC and c-Myc may serve as a target gene of Notch pathway signaling.
