Expression and significance of miR-214 in the bladder tissues of interstitial cystitis/bladder pain syndrome

摘要: 目的:探讨miR-214在间质性膀胱炎/膀胱疼痛综合征(IC/BPS)患者膀胱组织中的表达及意义。方法:30例IC/BPS患者,根据膀胱镜下表现,分为Hunner溃疡组(n=12)和点状出血组(n=18)。分别取病变组织和病变周围组织行实时定量逆转录聚合酶链反应(qRT-PCR)。分别比较两组中病变组织与其周围组织中miR-214的表达情况,以及两组间病变组织中miR-214的表达情况。结果:Hunner溃疡病变处组织miR-214表达水平0.49~1.23,平均(0.89±0.20),Hunner溃疡病变周围组织miR-214表达水平2.78~4.81,平均(3.78±0.63)。点状出血处病变组织miR-214表达水平0.94~1.84,平均(1.47±1.33),点状出血病变周围组织miR-214表达水平2.76~3.95,平均(3.60±0.42)。Hunner溃疡病变组织miR-214表达明显低于其周边组织。差异有统计学意义(t=18.76,P<0.000 1)。点状出血处病变组织miR-214表达明显低于其周边组织,差异有统计学意义(t=17.30,P<0.000 1)。Hunner溃疡病变组织miR-214表达明显低于点状出血处病变组织,差异有统计学意义(t=5.554,P<0.000 1)。结论:IC/BPS患者膀胱病变组织处较其周围组织miR-214表达明显降低,Hunner溃疡病变组织miR-214表达明显低于点状出血处病变组织。这可能与疾病的发生、发展有关。Abstract: Objective: To investigate the expression and significance of miR-214 in the bladder tissues of patients with interstitial cystitis/bladder pain syndrome(IC/BPS).Methods: Thirty IC/BPS patients were divided into Hunner's ulcer group(n=12) and punctate hemorrhage group(n=18) according to cystoscopy. Real-time quantitative reverse transcription polymerase chain reaction(qRT-PCR) was performed on the diseased tissues and the surrounding tissues respectively. The expression of miR-214 in the diseased tissues and surrounding tissues of the two groups was compared, and the expression of miR-214 in the diseased tissues between the two groups was also compared.Results: The expression level of miR-214 in the tissue of Hunner's ulcer was 0.89±0.20(0.49-1.23), and the expression level of miR-214 in the surrounding tissue of Hunner's ulcer was 3.78±0.63(2.78-4.81). The expression level of miR-214 was 1.47±1.33(0.94-1.84) in the lesions with punctate hemorrhage, and 3.60±0.42(2.76-3.95) in the surrounding tissues with punctate hemorrhage. The expression of miR-214 in the Hunner's ulcer was significantly lower than that in the surrounding tissues. The difference was statistically significant(t=18.76, P<0.000 1). The expression of miR-214 in the tissues at the punctate hemorrhage was significantly lower than that in the peripheral tissues, and the difference was statistically significant(t=17.30, P<0.000 1). The expression of miR-214 in the tissue of Hunner's ulcer was significantly lower than that in the lesion of punctate hemorrhage, and the difference was statistically significant(t=5.554, P<0.000 1).Conclusion: The expression of miR-214 in the diseased tissues of IC/BPS patients was significantly lower than that in the surrounding tissues, and the expression of miR-214 in the diseased tissues of Hunner's ulcer was significantly lower than that in the diseased tissues of punctate hemorrhage. This may be related to the occurrence and development of the disease.
